Effect of Lipophilic Fraction from Panax ginseng on Phosphatidylinositol Breakdown in Rat Platelets Activated by Collagen

Abstract
We have studied the effect of a lipophilic fraction (LF) from Panax ginseng C.A. Meyer on phosphatidylinositol (PI) breakdown in platelets stimulated with collagen. When platelets (109/§¢) prelabeled with [C14]-arachidonic acid (20:4) were stimulated by collagen (20 §¶/§¢), [C14-20:4]-PI was decreased compared with that of the control, unstimulated platelets. However, [C14-20:4]-PI was increased by an exogenous LF (20 §¶/§¢). Phosphatidic acid (PA) and [C14-20:4]-metabolites, breakdown-product of phosphoinositides such as PI, phosphatidylinositol-4-monophosphate and phosphatidylinositol 4,5-bisphosphate, were increased in collagen-stimulated platelets. But both [C14-20:4]-PA and [C14-20:4]-metabolites were reversed to the control level by LF added in collagen-stimulated platelet suspension. These results suggested that LF inhibits the breakdown of [C14-20:4]-PI in collagen-stimulated platelets. Thus, our data demonstrates that LF has an antiplatelet effect by inhibiting PI-breakdown.
